My guess would be either the power supply or hard drive. I would back up my files right away in case it's the hard drive. Can you take the side off the tower and see if you can figure out exactly where the sound is coming from?

Could also be internal fan(s) grinding. I had that prob last year. Replaced fans and no more grinding sound.

Prepare to vacuum the heck out of the internals and I recommend vacuuming the tower fans, the CPU fans and heat sinks.
A grinding sound is also a warning to get a good backup strategy in place. It will be the best $100 piece of mind around.

That's a good point. Vacuuming out the inside of the tower is so often neglected and all the dust that builds up can cause major problems. Canned air can work well too, but I wouldn't recommend it if there is a ton of dust. It can be messy!

I thoroughly blow-out all the machines in my house about every quarter. The dust build-up will start to stick and act as an insulator keeping all the heat inside the components. Heat is the main enemy of electronic devices. Keep it clean and parts will last much longer.
